Synthesis Method:
In the synthesis of Melanotan II, the ε-amino group of lysine and the γ-carboxyl group of aspartic acid undergo orthogonal deprotection prior to a carbodiimide-mediated lactamization, yielding an intermediate. Coupling this intermediate with N-acetyl-norleucine produces Melanotan II. The entire process can be completed in 12 steps with an overall yield of 2.6%, achieving a product purity of over 90% without the need for preparative chromatography.
